  • Overview

    While anti-VEGF biologics have transformed outcomes in neovascular age-related macular degeneration (nAMD), real-world data continue to show that visual gains achieved in clinical trials are not consistently replicated in practice due to various reasons. As sustained-delivery tyrosine kinase inhibitors (TKIs) advance through late-stage development, retina specialists are confronted with complex mechanistic distinctions, extracellular ligand sequestration versus intracellular receptor inhibition, pan-VEGFR activity, IL-6 pathway modulation, and potential effects on vascular stability, that carry meaningful implications for durability, disease control, and patient selection. The pivotal trials evaluating these TKIs also differ in terms of study design, statistical frameworks, and study endpoints, and without structured guidance, clinicians risk making inappropriate cross-trial comparisons, misinterpreting statistical intent, or overgeneralizing durability signals. This educational activity has been designed to analyze the mechanistic differences between TKIs and their relation to clinically relevant decision points and the key differences in TKI trial designs in nAMD.
